Survival and replication of Escherichia coli O157:H7 inside the mice peritoneal macrophages BJM
Al-Mariri,Ayman.
The replication of Escherichia coli O157:H7 on the resident peritoneal macrophages of four mice strains (BALB/c, CD1, C57BL, and Swiss) has been investigated. Macrophagial bactericidal killing activity was estimated via studying their ability to internalize (gentamicin-protected) E. coli during 2, 4, 24, and 48 h assays. Host genetic background has been found to show no significant effect on the ability of resident peritoneal macrophages to kill E. coli O157:H7.

Systemic injection of an H4 receptor agonist induces a decrease in CREB and pCREB levels in the cerebellar vermis and prefrontal cortex in mice BJMBR
Fernandes,C.E.M.; Serafim,K.R.; Gianlorenço,A.C.L.; Mattioli,R..
Studies have shown that an injection with the histamine H4 receptor agonist VUF-8430 modulates emotional memory processes. In the present study, the aim was to verify if intraperitoneal (ip) injection of VUF-8430 (500 ng/kg) in mice affects the synthesis of proteins required for memory consolidation processes by activating the phosphorylation of CREB (pCREB) in classical structures linked to emotional memory (prefrontal cortex, amygdala, and hippocampus) and the cerebellar vermis, a structure that has also been recently implicated in emotional memory. The results obtained using western blot analysis demonstrated that VUF-8430 induced a decrease in CREB and pCREB levels in the cerebellar vermis and prefrontal cortex, suggesting that this dose impaired the...

Silva,Suellen Maria Albuquerque da; Silva Neto,Geraldo José da; Nascimento,Ivenis Raphael Cavalcante do; Viana,Max Denisson Maurício; Lima,Alyne Almeida de; Bezerra,Pedro Henrique Simões; Bastos,Maria Lysete de Assis; Moreira,Magna Suzana Alexandre; Campesatto,Eliane Aparecida.
ABSTRACT In view of the traditional use of Tabebuia aurea for treating pain and inflammation, the antinociceptive pharmacological potential of T. aurea ethanolic extracts (TAEE) was investigated through in vivo experimental models. First, the MTT assay was conducted to determine the potential cytotoxicity of the TAEEs. Afterwards, the acetic acid-induced writhing test and the formalin-, and glutamate-induced nociception tests were performed on Swiss adult mice treated with TAEEs (100 and 200 mg/kg doses, p.o.), or saline solution (control groups, 10mL/kg, p.o.), or standard drugs: dipyrone 40 mg/kg (p.o.), and morphine 5,7 mg/kg (i.p). In the MTT assay, none of the tested concentrations demonstrated signals of cytotoxicity. In the in vivo experimental...

The combination of Passiflora alata and Valeriana officinalis on memory tasks in mice: comparison with diazepam BABT
Slomp Junior,Helvo; Seniski,Gerusa; Cunha,Cláudio da; Audi,Elizabeth Aparecida; Andreatini,Roberto.
The effect of a compound combining Valeriana officinalis and Passiflora alata extracts was tested on two mouse memory models: habituation and step-through inhibitory avoidance. Diazepam (1.0 and 2.5 mg/kg) was used as a positive control. Acute diazepam (2.5 mg/kg) before training impaired the habituation and performance in the inhibitory avoidance. On the other hand, acute phytotherapeutic compound (40-160 mg/kg), also before the training session, did not alter mouse behavior in these models. Repeated (15 days) treatment with the compound also did not impair the habituation. At the doses used, no locomotor effect was found. Taken together, the results suggest that, contrary to diazepam, the anxiolytic Valeriana officinalis and Passiflora alata compound did...

Dias,R.S.; Carmo,L.S.; Heneine,L.G.D.; Rocha,P.H.; Barbosa,C.F.; Rodrigues,R.J.; Linardi,V.R..
Acute toxicity test (LD-50) using toxic shock syndrome toxin (TSST-1) was tested in BALB/c, C57BL/6 and Swiss mice. Animals (n = 10) were intraperitoneally injected with TSST-1 (0.01-10.0µg/mouse) followed 4h later by potentiating dose of lipopolysaccharide (75.0µg of LPS - E. coli O111:B4) and cumulative mortality was recorded over 72h. Control animals received either TSST-1 or LPS alone. The data were submitted to qui-Square test and acute toxicity test was calculated by probit analysis (confidence limits expressed as µg toxin/kg). BALB/c mice was the most sensitive (20.0µg/kg, 95% confidence limits: 9.0-92.0) followed by C57BL/6 (38.5µg/kg, 95% confidence limits: 9.11- 401.6). Data from Swiss mice was not conclusive, indicating only low sensitivity....

TNF-α and IL-6 immunohistochemistry in rat renal tissue experimentaly infected with Leptospira interrogans serovar Canicola J. Venom. Anim. Toxins incl. Trop. Dis.
Marinho,M; Monteiro,CMR; Peiró,JR; Machado,GF; Oliveira-Júnior,IS.
Leptospirosis is a public health problem worldwide and its etiology remains unclear. Its pathogenesis involves a complex interaction between host and infecting microorganism. The inflammatory reaction that controls the infection process also underscores many pathophysiological events occurring in leptospirosis. We investigated the presence of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in renal tissues by immunohistochemical and histopathological examination in animals experimentally inoculated with Leptospira serovar Canicola. All the tests were carried out 2, 7, 14, 21 or 28 days after inoculation. Although TNF-α and IL-6 had been detected in tissues throughout the observation period, these cytokines appeared more intensely during the...

Treatment of critical defects produced in calvaria of mice with mesenchymal stem cells Anais da ABC (AABC)
Monteiro,Betânia S.; Argôlo-Neto,Napoleão M.; Nardi,Nance B.; Chagastelles,Pedro C.; Carvalho,Pablo H.; Bonfá,Laila P.; Filgueiras,Richard R.; Reis,Amanda S.; Del Carlo,Ricardo J..
Mesenchymal stem cells (MSC) are present in specialized niches in perivascular regions of adult tissues and are able to differentiate into various cell types, such as those committed to repairing. Bone marrow derived MSC from eight young mice C57BL/ 6 gfp+ were expanded in culture for repairing critical defects in calvarial bone produced in twenty-four young isogenic adult C57BL/6 mice. The animals were subjected to a cranial defect of 6.0mm diameter and divided into two equal experimental groups. Control group did not receive any treatment and the treated group received a MSC pellet containing 1.0 x 10(7) cells/mL into the defects. The group treated with MSC showed increased angiogenesis and amount of new bone deposited on the defect limits than that...

Ultrastructural Aspects of Spermatids in Isogenic Black Mouse C57BL6J International Journal of Morphology
Fernandes Boaro Martins,Márcia Regina; Carvalho Pinto e Silva,José Ricardo de.
The objective of this study was to describe some changes in spermatids differentiating events during spermiogenesis, leading to spermatozoa final formation. The animals were sacrificed with ethylic acid inhalation and, then, testis fragments were collected, fixed in Karnovsky and post-fixed in osmium tetroxide. After dehydration, the sections were stained with uranyl acetate 2%. It was observed in early spermatids at Golgi phase, formation of acrosomal granule contained in the acrosomal vesicle, spheroidal nucleus with rugous reticulum around it, and Golgi apparatus made up of lamellae and vesicular structures. At cephalic hood phase, the granule suffers flattening and the marginal fossa is formed. In both phases, it was possible to observe the presence of...

Ultrastructural Study of the Neuromuscular Junctions of Extensor Digitorum Longus Muscle (M. extensor digitorum longus) of the Mice International Journal of Morphology
Boaro,Sueli N; Fragoso Neto,Raul A.
The neuromuscular junction of the extensor digitorum longus muscle of fingers was analyzed in 21 young (three months) and old (from six to 25 months) mice, from both genders. Morphologic changes were found throughout the mouse life, being more frequent and visible with aging. According with the data described in the literature consulted and the observations taken in this research, it becomes clear that a continuous process of morphological remodeling occurs in all neuromuscular ultrastructural junctions of the extensor digitorum longus muscle of fingers, during the life of the animal. Theses changes are characterized by figures of myelin in the cytoplasm of Schwann cells, pleomorphic and multivesiclar bodies, mitochondrias with morphologically altered...
